Tafazolli, 34, is unable to play for four weeks after seeing a specialist for a broken elbow, while Kilkenny, 26, is out for eight weeks in what manager Ian Holloway said was "bitterly bad news".
"Taf disagrees with it but you can't argue with an X-ray," Holloway told BBC Radio Wiltshire.
"Gav's going to be out eight weeks, if there was eight weeks left. Joel McGregor tackled him - they were on the same team, I've never seen anything like it."
Swindon host Accrington Stanley on Saturday as they try to get their promotion push back on track following a loss to Colchester in their last outing, which left them fifth in the table.
The Robins have however welcomed on-loan Leicester defender Tom Wilson-Brown, who has not featured since February, and midfielder Joe Snowdon back into the fold - and they could feature on Saturday in some form.
"It's fantastic to have [Tom]. He's had two hamstring injuries since he's been here and the quality that he's got, he will definitely add to the group," Holloway said.
"The size factor and attacking the ball that we're losing with Taf's skillset, Tom will pick up on and be a great help if and when needed."
Swindon, who are four points behind Cambridge in third, also play Grimsby and play-off rivals Chesterfield in their final matches of the regular campaign.

Holloway described their lacklustre loss to Colchester as "nowhere near good enough" but added the players are looking to rebound quickly.
"The lads have trained well this week, not showed any disappointment. We're going to try and bounce back and I want to get a bit of freedom in their play, enjoyment in their play," he said.
Holloway said he hopes to be on the touchline on Saturday and is appealing against the red card he was shown in the 83rd minute of the 3-0 loss.
"I've talked to the League Managers Association about it and it's between me, them and the powers that be," he said.
