Chasing a daunting 229, Mumbai rode on sensational half-centuries from Rohit Sharma and Ryan Rickelton to complete the chase in just 18.4 overs.
Rohit smashed 84 off 44 balls, while Rickelton blazed 83 off only 32 deliveries. The duo stitched together a match-defining 143-run opening partnership, giving MI complete control of the chase from the outset.
The victory helped Mumbai register only their third win in 10 matches this season. They now move to six points with a net run rate of -0.649, remaining ninth in the standings but still mathematically alive in the playoff race.
Lucknow Super Giants, meanwhile, stayed rooted to the bottom of the table with just four points from nine matches and an NRR of -1.076.
Earlier, LSG finally showed signs of life with the bat after a disappointing campaign. Nicholas Pooran smashed his first fifty of the season, hammering 63 off just 21 balls in a brutal display of power-hitting. Mitchell Marsh chipped in with 44, while Himmat Singh (40 not out) and Aiden Markram (31 not out) provided the late flourish to push LSG to 228/5.
For MI, Corbin Bosch stood out with figures of 2/20.
At the top of the table, Punjab Kings continue to lead with 13 points from nine matches, followed by Royal Challengers Bengaluru on 12 points with the best net run rate among the top teams.
